That single fact does most of the work, so it's worth unpacking slowly — because almost everything people find confusing about NR comes from skipping over what it actually is and jumping straight to what it's marketed to do.
NR is a form of vitamin B3
Vitamin B3 isn't one molecule — it's a small family. Niacin (nicotinic acid) and nicotinamide are the two forms you'll see on most nutrition labels. Nicotinamide riboside is a third, more recently characterised member of that same family: nicotinamide joined to a ribose sugar.
Grouping NR under vitamin B3 isn't a technicality. In the EU it's the part that has real, official meaning. Vitamin B3 (niacin) carries a set of authorised European health claims describing its established role in the body — that it contributes to normal energy-yielding metabolism, to the reduction of tiredness and fatigue, to normal functioning of the nervous system, to normal psychological function, and to the maintenance of normal skin.6 Those claims apply when a product supplies a meaningful amount of the vitamin (at least 15% of niacin's Nutrient Reference Value per daily serving).
Hold onto that distinction, because it's the honest line running through this whole topic: the vitamin B3 story is settled and officially recognised. The NAD⁺-and-longevity story is a different, newer, and far less settled conversation.
How your body turns NR into NAD⁺
NAD⁺ (nicotinamide adenine dinucleotide) is a coenzyme involved in hundreds of reactions — most famously the ones that convert food into usable cellular energy. Your body makes NAD⁺ continuously, recycling it through what's called the salvage pathway, and it can also build it from dietary B3 precursors like NR.
When you take NR, enzymes phosphorylate it into NMN, and NMN is then converted onward into NAD⁺. It feeds the same NAD⁺ machinery your cells already run.
Research consistently reports that NAD⁺ availability declines with age. That observation is the entire engine behind the NAD⁺ supplement category: the reasoning goes that if levels fall, topping them back up might help. Whether that reasoning holds up in humans is exactly the question the evidence section below gets into — honestly.
NR vs NMN — and why NR is the one you can legally buy in the EU
This is the comparison most people are actually searching for, and there's now a clean answer to part of it.
Chemically, NMN (nicotinamide mononucleotide) is NR plus a phosphate group — one enzymatic step closer to NAD⁺. That “one step closer” is the basis for a lot of marketing implying NMN must therefore be superior. In practice, both molecules feed the same pathway, and until recently the head-to-head human data simply didn't exist.
In January 2026 it did. A Swiss randomised trial published in Nature Metabolism gave 65 healthy adults 1,000 mg/day of NR, NMN, nicotinamide, or placebo for 14 days and measured circulating NAD⁺ side by side. NR and NMN each raised NAD⁺ by roughly the same amount, with no significant difference between them; plain nicotinamide produced only a brief, transient effect.5 So on the one thing both are sold to do — raise NAD⁺ — NR and NMN came out comparable.
Here's the part that matters if you're shopping in Europe, and it has nothing to do with which molecule is “better”: NR is authorised for sale as a novel food in the EU. NMN is not. That means an EU shopper choosing an NAD⁺ precursor isn't really choosing between two equivalent options on the shelf — NR is the one that's legally sold here as a food supplement, while NMN sits in a regulatory grey zone. It's an unglamorous reason to prefer NR, but for a European buyer it's the decisive one.
What the human evidence actually shows
Now the honest part, kept in proportion.
Well established: NR reliably raises blood NAD⁺ in a dose-dependent way. The first human pharmacokinetic study found measurable increases after single 100, 300, and 1,000 mg doses.1 A longer randomised, placebo-controlled trial confirmed a sustained rise over eight weeks and found NR well tolerated at the doses tested.2 The 2026 head-to-head above adds more confirmation.5 On the biomarker, the case is strong.
Much less settled: whether raising that biomarker produces the health outcomes people are hoping for. Several rigorous trials have raised NAD⁺ successfully and then not found the downstream benefit — for example, a randomised trial in men saw no improvement in insulin sensitivity,3 and a well-designed twin study raised NAD⁺ without a corresponding improvement in metabolic health.4 That doesn't mean NR “doesn't work” — it means the link between “NAD⁺ went up” and “you're measurably better off” is still being established, and honest sources say so. Anyone promising more than that is ahead of the data.
What can — and can't — be said about NR's benefits
If you search “NR supplement benefits,” this is the useful answer: there are two different registers, and it's worth not mixing them.
Because NR is a form of vitamin B3, the officially recognised benefits are the vitamin B3 (niacin) claims — its contribution to normal energy-yielding metabolism, to reducing tiredness and fatigue, and to normal nervous-system and psychological function — where a product delivers enough of the vitamin to qualify.6 Those are the statements grounded in established science and EU authorisation.
The NAD⁺-longevity claims — reversing ageing, extending lifespan, and the rest — are a separate category, and they are not established or authorised. Treating the B3 register as the honest floor, and the NAD⁺ register as active science rather than a promise, is the accurate way to think about it.
Dosing and safety, briefly
Human trials have used NR across a wide range, commonly 100–1,000 mg per day, with some studies going higher. Its EU novel-food authorisation sets conditions of use, including a maximum intake for the general adult population and restrictions for certain groups such as pregnant and breastfeeding women.6 This isn't medical advice — if you take medication or have a health condition, that's a conversation for a clinician, not a supplement label.
The bottom line
NR is a form of vitamin B3 and a genuine NAD⁺ precursor. It reliably raises NAD⁺, it's well tolerated at studied doses, and — uniquely among the popular NAD⁺ precursors — it's authorised for sale in the EU. What it doesfor your body, in claim terms, is what vitamin B3 does. The bigger longevity story is real science that's still being worked out, and the honest version of this page would rather tell you that than sell you the shortcut.
